Methods excerpt · pooled screen + scRNA-seq
Abstract
Cellular stress responses emerge from coordinated regulatory programmes. We combined perturbation screening with transcriptomic profiling to map the pathways that shape resilient cell states.
Evidence source
Methods
A genome-scale CRISPR guide library was introduced across three biological replicates. Cell states were profiled by single-cell RNA sequencing and analysed in a reproducible pipeline.
Introduction
Previous studies identified individual regulators, but their combined influence across heterogeneous populations remained unresolved.
Results
Perturbation profiles separated adaptive and sensitive states across the sampled populations.
Discussion
These findings provide a framework for prioritising regulators and reproducible follow-up experiments.
